Step-by-Step Instructions
1. Prepare Your Workspace
• Disinfect your work surface.
• Work near a flame, in a still air box, or under a laminar flow hood to reduce contamination.
• Wash your hands and wear gloves if needed.
2. Label the Agar Plate
• Write the date and sample name on the bottom (agar side) of the plate before opening it.
3. Open the Swab Package
• Peel open the swab pack carefully, without touching the swab tip.
• Hold the swab by the handle only.
4. Collect the Sample
• Gently touch the swab to the surface or substance you want to transfer (e.g., spore print, mushroom cap, or culture surface).
• Roll or lightly drag the tip to pick up material.
5. Inoculate the Agar Plate
• Open the petri dish just enough to insert the swab (keep the lid hovering to prevent airborne contamination).
• Lightly streak the swab across the surface of the agar in a zig-zag pattern (or streak in sections if isolating).
• You can streak multiple areas if desired using different parts of the swab.
6. Seal and Incubate
• Close the petri dish and optionally seal it with parafilm or tape.
• Incubate at the appropriate temperature (usually 24–27°C for most fungi).
7. Clean Up
• Dispose of the swab properly.
• Disinfect your work area again.
